Re: Toxicity Analysis of Sediment Samples

Please find the enclosed reports for sediment toxicity testing conducted on samples received May 10, 1999. The reports include both the results and data regarding the test conditions.

As reported previously, tests conducted using Chironomus riparius  and initiated on June 8, 1999 were invalid because control mortality exceeded the maximum allowed under Environment Canada Protocols. As required under internal quality control practices, and investigation of the test system was initiated to determine if the cause of the control mortality could be identified. Our investigation included a review of internal procedures related to the performance of the test systems during the period. They included but were not limited to light intensity and photoperiod maintenance, temperature, feeding regime, aeration rates, preparation of test chambers, routine chemistry of control water, organism history and handling. No abnormalities were identified with respect to the operation of the test system. There was no unusual mortality observed in reference toxicant testing conducted within a week of the test. It must be noted however, that reference toxicant assays for sediment tests differ slightly from the actual sediment test in that they are shorter term and test performance of the organism is in a "water only" environment.

Remedial action included immediately informing the test sponsor, Golder Associates. Approval was given for a retest of the samples. The retest was initiated on June 23, 1999. Test reports for these sediments indicate that storage time exceeded 6 weeks.

Test Organisms:     No organisms exhibiting unusual appearance, behaviour, or undergoing unusual treatment were used in the test. Previous 48 h culture mortality <1%.

Control Sediment:     Obtained from Long Point, Ontario.

Test Conditions:     Sediment MR-R3 contained abundant small amphipods which could not be eliminated by hand picking. Sediment sample was sieved using a 500µm stainless steel mesh prior to testing. There were no other unusual conditions or deviations from the test protocol.
